Transient expression of N-Met-FLAG-tagged KIAA1401 in SKOV3 ovarian cancer cells. 48 hr. post-transfection, M2 anti-FLAG primary, Alexa488 chicken anti-mouse secondary,TRITC-phalloidin counterstain, confocal microscopy. KIAA1401 localizes to the nucleus, with nucleolar exclusion. Transient expression of N-Met-FLAG-tagged KIAA0732 in SKOV3 ovarian cancer cells. 48 hr. post-transfection, M2 anti-FLAG primary, Alexa488 chicken anti-mouse secondary, TRITC-phalloidin counterstain, confocal microscopy. KIAA0732 localizes to the cytoplasm.

Dr. Jan Ryerse will assume oversight of the confocal scanning microscope facility in the Department of Pathology.

The confocal imaging facility provides two types of instruments for viewing fluorescent specimens, a Perkins Elmer system which images living cells and tissues in real time and a BioRad system which images fixed and stained cell cultures and tissue sections. Both microscopes utilize filter sets for green-emitting fluorophores such as fluorescein and red-emitting fluorophores such as rhodamine.

As Director of the facility, Dr. Ryerse will train new users and provide advice and assistance in the design and completion of projects. Ms. Alice Rickard and Mr. Jeremy Grojean will assist Dr. Ryerse with scheduling, training and troubleshooting user issues.

The BioRad is available now for use by trained users. Contact Alice Rickard (office telephone 314-268-5220, email: rickarda@slu.edu ) to schedule usage time or sign up on the schedule posted on the BioRad room door (Room 1120, 11th Floor, University Hospital). The scope will be turned on and ready to go when you arrive. In the near future, we plan to have a sign-up site accessable via the Pathology Department’s website where trained users can schedule scope time online. New users should contact Dr. Ryerse (office telephone 314-977-7848, email: ryersejs@slu.edu ) to schedule a training session.

The Perkins-Elmer requires maintenance prior to being made available for general use but it should be up and running within the next few weeks. Contact Dr. Ryerse to schedule usage of the Perkin-Elmer system.

A charge of $30.00 per hour will be assessed for both confocal microscopes in order to defray the costs of the service contracts. However, to accommodate users who have samples which can be evaluated in a shorter time period or require an extended time period, billing will be in one-half hour increments.
